AI designs novel metalloproteases
Scientists have utilized RFdiffusion to create novel zinc-based metalloproteases. These designed enzymes feature active sites that activate water near peptide substrate binding areas. The research demonstrated that 36% of the designed enzymes effectively cleaved their target peptide substrates, with one design achieving more than 100 million times faster catalysis. This computational method was also successful in engineering proteases that specifically break down disease-related proteins like TDP-43, amyloid-beta, and serum amyloid A.
